Basic Principle of Bisulfite Conversion
The principle of bisulfite conversion is based on the chemical conversion of unmethylated cytosine to uracil, while methylated cytosine is exempt from this conversion, allowing DNA methylation to be determined at the single-base resolution. Bisulfite conversion is a process in which DNA is denatured (made into a single strand) and treated with sodium bisulfite, resulting in deamination of unmethylated cytosine to uracil, while methylated cytosine (5-mC and 5-hmC) remains unchanged.
