| Scientific Background: | SMYD3 (SET and MYND domain containing 3) is a member of the SMYD family of protein methyltransferases. All family members (SMYD1, SMYD2, SMYD3, SMYD4, and SMYD5) contain a conserved catalytic SET domain, first identified in Drosophila Su[var]3-9, Enhancer of zeste, and Trithorax proteins. SMYD3 can be overexpressed in several different cancers, including breast, colorectal, and liver cancer. Research shows that cancer cells may express full-length and a more active cleaved SMYD3 that lack the first 34 amino acids at the N-terminus. The N-terminus of full-length of this gene interacts with Hsp90alpha and enhances its activity. This indicates that the N-terminal region may participate in regulating methyltransferase activity. |
