| Background: | PRDM5 (PR Domain Containing 5) is a transcription factor of the PR-domain protein family. It contains a PR-domain and multiple zinc finger motifs. Mutations of PRDM5 causes Type 2 brittle cornea syndrome (BCS2) which is an inherited connective tissue disease with a devastating ocular phenotype through histone and DNA methylation. PRDM5 is a relevant tumour suppressor gene that is frequently targeted in colorectal tumourigenesis. Promoter methylation-mediated downregulation of PRDM5 contributes to the development of lung squamous cell carcinoma. It is a sequence-specific DNA-binding transcription factor and represses transcription at least in part by recruitment of the histone methyltransferase EHMT2/G9A and histone deacetylases such as HDAC1. |
