| Description: | Protein arginine methyltransferases (PRMT) are a family of enzymes with highly conserved catalytic domains that catalyze the transfer of a methyl group from S-adenosyl-L-methionine to a specific arginine residue in a target protein. PRMT4, also known as coactivator-associated arginine methyltransferase 1 (CARM1), is a type-1 PRMT, catalyzing ω-NG-monomethylarginine (MMA) and ω-NG,NG-dimethylarginine (aDMA) on histone H3, Arg-17 and Arg-26. PRMT4/CARM1 acts as a transcriptional coactivator by binding the p160 coactivator complex (SRC-1, GRIP1/TIF2, and pCIP/RAC3/ACTR/AIB1/TRAM1) thereby enhancing nuclear receptor function. PRMT4/CARM1 activity is inhibited by phosphorylation at Ser217, which prevents binding of S-adenosylmethionine. PRMT4 is ubiquitously expressed and is thought to play an important role in hormone mediated transcriptional regulation and lipid metabolism. |
